Matching the Block to the Tube
The block well should hold the tube so that the sample volume sits fully inside the heated section, with the tube standing vertically and the rim clear of the lid. A well that is too wide leaves an air gap and slows heat transfer; a well that is too tight makes tubes jam and crack. Where a method uses more than one tube size, order a block for each format rather than forcing adapters, and keep the extractor tool with the block so hot tubes can be removed safely.
Cover the block or close the heated lid whenever a method runs at high temperature for more than a few minutes. Without a cover the top of the tube stays cool, condensate forms on the wall and the concentration of the sample changes. This is the single easiest improvement to make to an existing incubation step.

Remember that the display reads the block, not the sample. After the unit reaches its set point, allow the tubes to equilibrate before starting the timer, and keep the tube type, fill volume and block position the same when a method is repeated. When you validate a method, measure the sample temperature rather than trusting the digital readout.
Dry Block versus Water Bath
A water bath has more thermal mass and better uniformity across many samples at once, which suits large volumes and long incubations at moderate temperatures. A dry block heater is faster to set up, cannot spill, keeps labels dry, takes up less bench space and works well above the boiling point of water. In practice, most Singapore laboratories keep both: the bath for bulk work and the block for tube-scale steps and anything above 100 degrees Celsius.
